There are always exceptions to rules of pronunciation in English, but as a general rule, doubling an intervening consonant shortens the immediately preceding vowel. So in "twining" the first "i" is pronounced line the "i" in "line", but in "twinning" it's pronounced like the "i" in "bit." It's one of the reasons I always write "targetting" instead of "targeting," even though the spell-checkers don't like it.
