The simple answer is that the well, for all intents and purposes, is a one-time use fixture.

The 'complete' answer is this.

When you attempt to drink from this well, the game evaluates two conditions;

(1) Is the Luck of the party member less than 15? If so, proceed. If not, no luck given ("Refreshing" result).

(2) Has the well been used before? If not, proceed. If it has been used before, no luck ("Refreshing" result)

I don't think that the well "respawns" with the other features of this map.
